- PROCEDURE:
- Connect the unit to the desired point of sample using sterile tubing. The inlet gas valve should be in a closed position.
- Insert an identified petri dish with sterile medium SCD in the “Pinocchio funnel” housing and screw it to the conical head.
- Open the line of Compressed air or gas line.
- Open the gas valve ‘D’ + ‘D1’ and adjust the airflow on flow meter to 100 Liters / min (210 SCFH) to wash the line.
- Connect the “Pinocchio funnel” to the “Pinocchio Super”.
- Close the ‘D1’ valve and open ‘D2’ to start the countdown.
- Close the gas valve after 10 minutes (1000 liters of air is collected).
- Unscrew the head and transfer the petri dish to the laboratory for incubation.
- The “Pinocchio Super” is ready for a new test.
- Incubate the plates in the inverted position at 30-35°C for NLT 72 hours.
- Count the colony forming units in the plate at the end of incubation time and report the result as cfu/1000 liters of gas/ compressed air.
- After incubation count the colonies. If any colonies are observed streak on selective media.
- Positive control and negative control for the respective media shall also be maintained.
LIMITS: 5 cfu/1000L; Pathogens: Enterobacteriaceae & Pseudomonas should be absent.
Frequency: Half yearly.
