OPERATION OF COLONY COUNTER (MAKE PROTOCOL)

OBJECTIVE:

To lay down the procedure for operation of colony counter (Make: Protocol).

SCOPE:

This SOP is applicable for the procedure for operation of colony counter (Make: Protocol) at {Company Name} {Location}.

RESPONSIBILITY:

  • In charge- Microbiology- is responsible to ensure compliance as per SOP.
  • Head/Designee Quality Control – Shall be responsible for ensuring compliance as per SOP.

ACCOUNTABILITY:

QA Head shall be Accountable for implementation of SOP.

What is a Colony Counter?

A colony counter is a laboratory instrument used to accurately count the number of bacterial or fungal colonies growing on an agar plate. It streamlines the counting process, reducing human error and saving time. Modern colony counters often use image analysis software to automatically detect and count colonies, further improving efficiency and accuracy. This tool is essential in microbiology research, food safety testing, and pharmaceutical development.

PROCEDURE:

Press the rear push button next to the power supply and switch on the monitor.

Now system is ready for counting the plate.

Place the plate under the camera.

The image of the plate will be grabbed automatically and displayed in the image pane.

If necessary, exclude any region within the frame where you do not want to count colonies, such as an area where colonies have coalesced and cannot be separated.

To perform a manual count, right-click in the image pane to display the image context menu and choose dialog box.

Count each colony by pressing with mouse on the displayed images on the screen.

After completing the count, the entry shall be made in Format-I.

Plate validation:

To validate the colony counter by using the standard validation plate.

Remove all dust from the standard validation plate.

Place the validation plate on the protocol unit in the protocol system in place of the dish holder.

Make sure that the colonies are all within the frame boundary.

Count the colonies on the validation plate by pressing the spacebar.

If the count per frame is 100, the system is working correctly.
