INTRODUCTION:

    According to Annex 15 of the EU-GMP Guide, Risk Assessment is a systematic process used to identify, analyze, evaluate, control and review potential risks associated with equipment, utilities, systems and manufacturing processes that may affect product quality, patient safety, data integrity and regulatory compliance.

    Failure Mode and Effects Analysis (FMEA) is one of the widely accepted Quality Risk Management (QRM) tools described in ICH Q9 (Quality Risk Management). It provides a structured methodology for identifying possible equipment failures, evaluating their impact on product quality and determining appropriate mitigation measures before qualification and routine operation.

    The Fluid Bed Dryer (FBD) is a critical equipment used in the Oral Solid Dosage (OSD) pharmaceutical manufacturing process. It is used for drying wet granules by passing controlled quantities of conditioned and heated air through the product bed. The process removes moisture from wet granules and achieves the specified moisture content required for subsequent manufacturing operations.

    The quality of dried granules directly depends upon the consistent performance of the Fluid Bed Dryer. Any malfunction may result in:

    • Inadequate drying
    • Over-drying
    • Non-uniform drying
    • Excessive product temperature
    • Residual moisture outside specification
    • Granule degradation
    • Change in physical properties of granules
    • Product contamination
    • Cross contamination
    • Filter failure
    • Improper airflow
    • Improper inlet air temperature
    • Improper exhaust air conditions
    • Product loss
    • Equipment breakdown
    • Batch rejection

    Therefore, a comprehensive Failure Mode and Effects Analysis (FMEA) is essential to identify all GMP-critical risks associated with the design, installation, operation, cleaning, maintenance and qualification of the Fluid Bed Dryer.

    This Risk Assessment forms the basis for Design Qualification (DQ), Installation Qualification (IQ), Operational Qualification (OQ) and Performance Qualification (PQ), ensuring that all applicable GMP requirements are adequately addressed prior to routine manufacturing.
