PROCEDURE FOR CALIBRATION OF POLARIMETER

  • PROCEDURE:
  • Make – Rudolph Research Analytical
  • Operate instrument as per operation SOP.
  • Sample compartment shall be kept empty while switching on instrument.
  • Switch on the instrument by using the ON/OFF switch present on backside of the instrument.
  • Keep the instrument ON condition at least for10 minutes before operation and wait until the reading on the display come to stable position.
  • Press the Zero key and wait until the display shows 0.00 reading.
  • Temp Trol Quartz control plate shall be placed right side in the trough.
  • Insert the built in temperature probe into the Temp Trol Quartz control plate.
  • Set the temperature to 20°C and allow it to stabilize.
  • Measure the optical rotation of the Quartz control plate at wavelength mentioned below.

Note: To the above acceptance limits add instrument Accuracy ± 0.01.

  • Temperature Calibration:
  • Select temperature calibration by pressing the temperature calibration key.
  • Place the temperature validation cell into the trough on to the right hand side of the trough.
  • Be sure the foot of Temp trol cell is clean and sitting flat against the clean temp trol temperature transmission surface to allow for maximum heating and cooling efficiency.
  • The built in temperature probe shall be placed in the temperature probe well of the temperature validation cell and tighten the finger nut to a secure.
  • Fill the temperature validation cell with water by adding the water into the temperature validation cell.
  • Fill the cell until the water level is just above the copper sleeve inside the thermometer well.
  • Insert a calibrated thermometer into the thermometer well of the temperature validation cell.
  • Set the temperature to 20°C and allow the built in probe temperature reading to stabilize at 20°C ± 0.1°C for 10 minutes and note down the temperature of the calibrated thermometer.
  • The same procedure shall be followed for 25°C and 30°C respectively and note down the reading.
  • Acceptance criteria shall be ± 0.2°C
  • Enter the details of the usage in the User logbook.
