INTRODUCTION

HVAC (Heating, Ventilation and Air Conditioning) systems are critical utilities in an Oral Solid Dosage (OSD) pharmaceutical manufacturing facility. The HVAC system provides controlled environmental conditions required for manufacturing, dispensing, sampling, compression, coating, packing and other controlled areas.

The HVAC system is responsible for maintaining appropriate:

  • Temperature
  • Relative Humidity (RH)
  • Air cleanliness
  • Air changes
  • Differential pressure
  • Airflow direction
  • Filtration
  • Air distribution
  • Pressure cascade
  • Prevention of cross-contamination
  • Environmental control

Failure of the HVAC system may adversely affect product quality, process performance, personnel safety and GMP compliance.

Potential consequences of HVAC failure include:

  • Temperature excursion
  • Relative humidity excursion
  • Loss of differential pressure
  • Incorrect airflow direction
  • Increased particulate contamination
  • Microbial contamination risk
  • Cross-contamination between products
  • Dust migration
  • Failure of containment
  • Product quality deterioration
  • Impact on granulation and drying processes
  • Tablet compression problems
  • Coating defects
  • Increased sticking/picking during compression
  • Packaging material or product deterioration
  • Failure of environmental conditions specified for manufacturing
  • Regulatory non-compliance
  • Production interruption
  • Batch rejection or investigation
  • Potential product recall in severe cases

This Failure Mode and Effects Analysis (FMEA) has therefore been prepared to systematically identify, evaluate and control risks associated with failure of the HVAC system at the OSD pharmaceutical manufacturing facility of AABBCC.

The assessment is prepared following the Quality Risk Management principles described in the attached reference report and is aligned with the general principles of WHO GMP, EU GMP, ICH Q9 and pharmaceutical facility requirements.
