  1. INTRODUCTION:

According to the definition given in Annex 15 to the EU-GMP Guide, a risk assessment is a method to assess and characterize the critical parameters in the functionality of an equipment or a process. Therefore, risk assessment is a key element in the qualification and validation approach.

In the project context, risk assessment for the dispensing booth / RLAF dispensing booth is performed as a basic GMP / EHS risk assessment to identify important GMP, operational, and EHS requirements.

The assessment considers the risks associated with dispensing of pharmaceutical raw materials, particularly the prevention of contamination, cross-contamination, powder escape, inadequate airflow, incorrect pressure differential, ineffective filtration, improper cleaning, and personnel exposure.

The structure and approach are based on the attached Vibro Sifter assessment, which identifies risks according to GMP and non-GMP categories and evaluates them using qualitative likelihood and consequence criteria.

  • OBJECTIVE OF RISK ASSESSMENT

At the basic stage of design, the risk assessment is carried out to verify that all features are taken into consideration to avoid the risk of failure of critical GMP and EHS parameters of the dispensing booth.

During the study, all GMP, EHS and operational parameters are identified and assessed for risk. Appropriate mitigation measures are proposed, and verification points are identified and defined.

The risk assessment report provides documented evidence that the design concepts and requirements are complete in considering relevant GMP, EHS and operational risks.

The objectives are to:

  • Review the design around a structured methodology.
  • Identify failure modes and associated risks.
  • Identify risks related to dispensing operations.
  • Evaluate the adequacy of existing control measures.
  • Identify risks related to air handling and filtration.
  • Identify risks related to cross-contamination.
  • Identify risks related to personnel exposure.
  • Identify recommendations to obtain a more acceptable risk level where required.
