INTRODUCTION:

    According to Annex 15 of the EU-GMP Guide, Risk Assessment is a systematic process used to identify, analyze, evaluate, control and review potential risks associated with equipment, utilities, systems and manufacturing processes that may affect product quality, patient safety, data integrity and regulatory compliance.

    Failure Mode and Effects Analysis (FMEA) is one of the most widely accepted Quality Risk Management (QRM) tools described in ICH Q9 (Quality Risk Management). It provides a structured methodology for identifying possible equipment failures, evaluating their impact on product quality and determining appropriate mitigation measures before qualification and routine operation.

    The Auto Coating Machine is one of the most critical pieces of equipment used in the Oral Solid Dosage (OSD) manufacturing process. It is designed to apply a uniform film coating or sugar coating onto tablets by spraying coating solution onto a continuously tumbling tablet bed under controlled processing conditions.

    The quality of coated tablets directly depends upon the consistent performance of the Auto Coating Machine. Any malfunction may result in:

    • Non-uniform coating
    • Colour variation
    • Coating peeling
    • Orange peel effect
    • Picking
    • Twinning
    • Tablet sticking
    • Surface roughness
    • Mottling
    • Spray drying
    • Overwetting
    • Tablet breakage
    • Cross contamination
    • Product mix-up
    • Coating solution wastage
    • Batch rejection
    • Equipment breakdown
